Corydalis saxicola Bunting total alkaloids attenuate paclitaxel-induced peripheral neuropathy through PKC?/p38 MAPK/TRPV1 signaling pathway

Abstract Background Corydalis saxicola Bunting, affiliated with the Papaveraceae Juss., has been proven to work well in anti-inflammation, hemostasis, and analgesia. This study was designed observe effect potential mechanism of Bunting total alkaloids (CSBTA) on paclitaxel-induced peripheral neuropathy (PIPN). Materials methods Rats were injected 2 mg/kg paclitaxel 4 times administrated 30 or 120 CSBTA. Mechanical thermal allodynia hyperalgesia tested. After 40 days, serum collected detect PGE2, TNF-?, IL-1? by ELISA. The L4-L6 segment spinal cord, DRG, plantar skin harvested, Western-blot RT-qPCR analyzed protein gene levels pro-inflammatory cytokines, p38 MAPK, PKC?, TRPV1. PIPN cell model established (300 nM, 5 d) primary DRG neurons. We examined CSBTA (25 ?g/ml 50 ?g/ml) measuring mRNA TNF-? CGRP, expression PKC?/p38 MAPK/TRPV1 signaling pathway model. Results results showed that effectively ameliorated hyperalgesia, regulated cytokines' contents (PGE2, IL-1?) neuropeptides (CGRP SP) different tissues vivo. In addition, significantly decreased cytokine neurons CGRP) expressions vivo vitro. Conclusion Therefore, a perspective therapeutic treatment neuropathy.
